Lentinan Impairs the Early Development of Zebrafish Embryos, Possibly by Disrupting Glucose and Lipid Metabolism

LNT is the major biologically active substance extracted from Lentinus edodes (L. edodes). Although functional and pharmacological studies have demonstrated that LNT has multiple benefits for animals and humans, the safety assessment is far from sufficient. To evaluate the potential safety risk, larval zebrafish were continuously exposed to varying concentrations of LNT for 120 h. The 96 h LC50 of LNT was determined to be 1228 μg/mL, and morphological defects including short body length, reduced eye and swim bladder sizes and yolk sac edema were observed. In addition, LNT exposure significantly reduced the blood flow velocity and locomotor activity of larval zebrafish. The biochemical parameters were also affected, showing reduced glucose, triglyceride and cholesterol levels in zebrafish larvae after being exposed to LNT. This phenomenon has been implicated in the pathogenesis of several gastrointestinal (GI) diseases, including gastritis, constipation, and inflammatory bowel diseases, which include Crohn’s disease, ulcerative colitis, functional dyspepsia, acid reflux, diverticular disease, and irritable bowel syndrome. Exploring the potential usage of the acellular preparation of porcine hemoglobin (PHb) isolated from slaughterhouse blood as a cell culture media component, we have tested its effects on the functional characteristics of stromal cells of mesodermal origin. Human peripheral blood mesenchymal stromal cells (PB-MSCs) were used in this study as a primary cell model system, along with three mouse cell lines (ATDC5, MC3T3-E1, and 3T3-L1), which represent more uniform model systems. We investigated the effect of PHb at concentrations of 0.1, 1, and 10 μM on these cells’ proliferation, cycle, and clonogenic and migratory potential, and found that PHb’s effect depended on both the cell type and its concentration. In the current study, eleven sooty mold isolates were collected from different tropical host plants. The isolates were identified under Capnodium, Leptoxyphium and Trichomerium, based on morphology and phylogeny. For the secondary metabolite analysis, the isolates were grown on Potato Dextrose Broth (PDB). The well-grown mycelia were filtered and extracted over methanol (MeOH). The metabolites in the growth medium (or filtrate) were extracted over ethyl acetate (EtOAc). The COVID-19 pandemic has influenced the epidemiology of other respiratory pathogens, and this was most evident in the 2020−2021 season, which was characterized by a low circulation of influenza viruses. We aim to present a comparative analysis of clinical and epidemiological characteristics of 2018−2019 influenza cases and 2020−2021 COVID-19 cases, hospitalized at a tertiary infectious diseases hospital in Bucharest. We used data collected from patients admitted for severe acute respiratory infection (SARI) and subsequently confirmed with either influenza or COVID-19. During the 2018−2019 season, 208 patients over 18 years of age were confirmed with influenza (median age = 53 years, 59.6% were female) and 6.7% had been vaccinated against influenza. Cytokine oncostatin M (OSM) plays an important role in a variety of inflammatory reactions and is mainly produced in neutrophils in inflammatory diseases. The accumulation of various types of waste containing both organic and inorganic metal-containing compounds is extremely hazardous for living organisms. The possibility of polymer degradation, biohydrogen synthesis, and metal detoxification via the dark fermentation of model potato waste was investigated. For this purpose, the strict anaerobic strain was isolated and identified as Clostridium butyricum. The high efficiency of dark hydrogen fermentation of potatoes with yield of hydrogen in 85.8 ± 15.3 L kg−1 VSpotato was observed. The copperand chromium salts solutions were added to the culture fluid to obtain the concentrations of 50, 100, and 200 mg L−1 Cu(II) and Cr(VI) in the active phase of growth (19 h of cultivation). In this study, the inhibitory activities against human monoamine oxidases (hMAOs) were evaluated using a library of 195 endogenous lichen fungi from Ukraine. Among them, the extract ELF68 of the endogenous fungus Rosellinia corticium from the lichen Pseudevernia furfuracea (L.) Zopf. exhibited the strongest inhibitory activity against hMAO-A. Using the activity-guided method, (S)-5-methylmellein (5MM) was isolated from the extract and had an IC50 value of 5.31 µM for hMAO-A with a lower potency for hMAO-B (IC50 = 9.15 µM). Compound 5MM also moderately inhibited acetylcholinesterase (IC50 = 27.07 µM) but very weakly inhibited butyrylcholinesterase and β-secretase. Compound 5MM had a Ki value of 2.45 μM and was a reversible competitive inhibitor of hMAO-A. A molecular docking study predicted that (S)-5MM showed higher binding affinity for hMAO-A (−6.8 kcal/mol) than hMAO-B (−6.4 kcal/mol). Carbon Black Replacement in Natural Rubber Composites Using Dry-Milled Calcium Carbonate, Soy Protein, and Biochar

Recent discoveries have shown that calcium carbonate and soy protein interactions can be used to reinforce rubber composites with improvements on the effective crosslink density and moduli. However, the method to incorporate the soy protein into the rubber matrix may be costly to scale up, since it involves microfluidization and drying steps prior to rubber compounding. In this work, a simpler process involving dry-milled calcium carbonate and soy protein was used to explore filler blends of calcium carbonate, soy protein, biochar, and carbon black. By blending these filler materials in various ratios, rubber composite samples with 40−50% of the carbon black replaced by sustainable alternatives were made. These composites had essentially the same tensile strength, with better toughness and elongation properties relative to the carbon black control. These composites would reduce dependence on petroleum and be more amenable to the rubber composite compounding infrastructure.

The release of winery wastewater (WW) into the environment, without proper treatment, can cause severe problems to freshwater quality and natural fauna and flora. Acetone released into the atmosphere can adversely affect human health and the environment. The aim of this work was to evaluate the performance of a laboratory-scale biotrickling filter (BTF) with bioball packing material to remove acetone vapor from contaminated air. The acetone removal efficiency was investigated in two different scenarios: with and without the inoculation of microorganisms. Three strains of bacteria, Pseudomonas putida, Rhodococcus aerolatus, and Aquaspirillum annulus, were used in the BTF. In both cases, the filter units were simultaneously operated for 100 days under three different inlet acetone concentrations (0.18 ± 0.01 g/m3, 0.25 ± 0.01 g/m3, and 0.40 ± 0.02 g/m3) and two different gas flow rates (2.54 and 5.09 m3/h). Maqui (Aristotelia chilensis (Mol.) Stuntz) is a Chilean berry rich in antioxidants, which are mostly found in the pulp and skin of the fruit. The objective was to evaluate the cryoconcentration process by centrifugation−filtration as a simultaneous, efficient, and innovative method to increase the content of thermosensitive bioactive compounds of aqueous maqui extract. Cryoconcentration separated the concentrated solute from the aqueous maqui extract with an efficiency of more than 95%; it increased the content of total polyphenols and total anthocyanins and antioxidant capacity by 280%, 573%, and 226%, respectively. Although the concentrates obtained by evaporation at 50, 70, and 80 °C increased the content of bioactive compounds, they did so in a lower percentage than the cryoconcentrate. Biogenic amines (BAs) are considered a potential microbiological toxicological hazard in aged cheese. Risk mitigation strategies include good hygiene practice measures, thermal treatment of milk and the use of competitive dairy cultures. The aim of this study was to evaluate the amount of BAs—tryptamine, β-phenylethylamine, putrescine, cadaverine, histamine, tyramine, spermidine and spermine—in the core and rind of cheeses ripened by bacteria (n = 61) and by mold cultures (n = 8). The microbial communities were counted, and the dominant lactic acid bacteria (LAB) were identified, corresponding to the BA concentrations. The total BA content was highest in the core of semi-hard cheeses (353.98 mg/kg), followed by mold cheeses (248.99 mg/kg) and lowest in hard cheeses (157.38 mg/kg). The study presents a comparative investigation of the composition and bioactivity of oil extracted from pomegranate seeds of sun-burned fruit (SB) and healthy fruit (HF) for the value-adding potential of pomegranate fruit waste. Seeds from SB and HF were independently freeze dried, sun dried, and oven dried before ultrasound-assisted oil extraction using petroleum ether. The extracted oil was analysed for yield, refractive index, ρ-anisidine value, total phenolic content (TPC), DPPH radical scavenging ability, antimicrobial activity, tyrosinase enzyme inhibition ability, and fatty acid composition. The results showed that oven dried seeds, regardless of fruit quality, yielded the highest oil (20.85−24.70%, dry weight). This study aimed to evaluate the effects of partial substitution of sodium chloride (NaCl) with potassium chloride (KCl) in combination with high-pressure processing (HPP) on the physicochemical properties and volatile compounds of beef sausage during cold storage at 4 °C. Significant differences were found in the volatile compounds of beef sausages with 0%, 25%, and 50% NaCl contents partially substituted with KCl subjected to 28 days of storage and were well-visualized by heat map analysis. A total of 75 volatile compounds were identified and quantified in the beef sausages at the end of 28 days of storage, including 12 aldehydes, 4 phenols, 2 ketones, 18 alcohols, 8 acids, 3 esters, 14 terpenes, and 14 alkanes. Pyraclostrobin is a fungicide extensively used for the control of various fungal diseases and is frequently detected in environmental samples. Natural systems, such as constructed wetlands (CWs) and gravity filters, are effective and environmentally friendly treatment systems, which can reduce or eliminate pesticides from the environment. Dynamics of the microbiological indicators of fresh sludge from wastewater treatment plants with a concentration of CaO, 10% and 20%, and ash, −30% and 50%, and treated with quicklime, ash, and microbial fertilizer for a 50-day period of composting were studied. The influence of temperature, water content, and oxygen on the development of microbes was established in laboratory conditions. Microbiological analysis included the determination of non-pathogenic (non-spore-forming bacteria, bacilli, actinomycetes, micromycetes, bacteria digesting mineral nitrogen), and pathogenic (Salmonella, Listeria, Escherichia coli, Enterococcus, Clostridium perfringens) microorganisms.
