February 21, 2023 (v1)
Subject: Biosystems
Keywords: anti-bacterial activity, anti-cancer activity, chemical composition, Cinnamomum verum, essential oil, Vietnam
(Cinnamomum genus) essential oil is commonly used in food preparation and traditional medicines, with a broad spectrum of biological activities. However, research on the extraction of essential oils (EOs) from C. verum cultivated locally in Vietnam is currently limited. Therefore, in this study, the chemical compositions and bioactivities of EOs from the bark and leaves of C. verum collected from the Thai Nguyen and Yen Bai provinces of Vietnam were investigated. The EOs samples were extracted by using water distillation, organic solvent (n-hexane) and ultrasound-assisted (in n-hexane solvent) extraction methods. The chemical composition of the obtained EOs were analyzed by GC-FID and GC/MS analyses. Results showed that the major chemical compositions of C. verum EOs were: (E)-cinnamaldehyde, trans-cinnamic acid, cinnamyl acetate, and benzaldehyde. Furthermore, C. verum EOs exhibited inhibitory activities against two tested cancer cell lines and four bacterial strains. These findings p... [more]

Subject: Biosystems
Keywords: Bojungikki-Tang, immunotherapy, KLN205, lung cancer, PD-1, tumor microenvironment
Although immunotherapy has become the standard of treatment for non-small cell lung cancer (NSCLC), only a limited number of patients benefit from it clinically because of the resistance to immunotherapeutic agents. Of late, the usefulness of herbal medicines in combination with immunotherapy has been investigated. Bojungikki-Tang (BJIKT) is a widely used traditional herbal medicine. It synergistically enhances the antitumor effects of chemotherapy and regulates the immune responses in cancer, but its antitumor effect with immunotherapy in NSCLC is unclear. In this study, we investigated the combined effects of BJIKT and an anti-PD-1 antibody in a KLN205-DBA/2 syngeneic lung cancer model. Immunohistochemistry and flow cytometry analyses were performed to analyze the changes in immune cells in the tumor microenvironment. BJIKT plus an anti-PD-1 antibody treatment significantly inhibited tumor growth, unlike the respective monotherapies. Compared to monotherapy, the combination treatment... [more]

Expression Analysis of BIRC3 as One Target Gene of Transcription Factor NF-κB for Esophageal Cancer
February 21, 2023 (v1)
Subject: Biosystems
Keywords: BIRC3, cytokine, esophageal cancer, transcription factor, tumor necrosis factor α (TNFα)
Esophageal cancer (ESCA) is one of the highest lethal malignancy tumors worldwide. Baculoviral IAP repeat-containing protein 3 (BIRC3) is the main inhibitor of apoptosis in many malignancies. The aim of this study was to clarify how BIRC3 acts in ESCA cells. Through TNMplot and GEPIA2 analysis, BIRC3 was found abundantly expressed in ESCA cells. The quantitative RT-PCR assay confirmed BIRC3 was pronouncedly induced in all used ESCA cell lines. In addition, proinflammatory cytokines TNFα and IL-1β were shown to have promotion effects on BIRC3 expression in ESCA cells. These promotive effects were blocked when the function of NF-κB was inhibited by bay 11-7082, which indicates the expression of the BIRC3 gene was regulated via the NF-κB transcription pathway in ESCA. Moreover, bioinformatics analysis showed that the BIRC3 gene had many NF-κB binding cis-elements. Chromatin immunoprecipitation was then performed and it was found that NF-κB directly interacts with cis-elements of the... [more]

Study on the Disinfection Efficiency of the Combined Process of Ultraviolet and Sodium Hypochlorite on the Secondary Effluent of the Sewage Treatment Plant
February 21, 2023 (v1)
Subject: Biosystems
Keywords: combined disinfection, disinfection efficiency, fecal coliform, secondary effluent, sodium hypochlorite, ultraviolet
The combined disinfection process of ultraviolet and sodium hypochlorite has more advantages than the single disinfection method in reducing the disinfectant dosage, shortening the reaction time, and resisting the impact of water quality changes and inhibiting the light reactivation of microorganisms. Given this, using the secondary effluent of a sewage plant as the research object, the disinfection efficiency of the combined process of ultraviolet and sodium hypochlorite was investigated. The experimental results showed that the inactivation effect of UV followed by sodium hypochlorite on fecal coliform and the inhibition of microbial photoreactivation was more significant than that of simultaneous disinfection of UV and sodium hypochlorite disinfection. When the UV dose was 24 mJ/cm2, after disinfection with UV followed by sodium hypochlorite, only 1 mg/L of sodium hypochlorite was required to be added, and a contact reaction time of 1 min for the fecal coliform index to meet the fir... February 21, 2023 (v1)
Subject: Biosystems
Keywords: biosynthesis, Reichstein process, two-step fermentation process, vitamin C
Vitamin C, C6H8O6, is a water-soluble vitamin that is widespread in nature. It is an essential nutrient involved in many biological processes in the living organisms: it enhances collagen biosynthesis, ensures the optimal functioning of enzymes and the immune system, has a major role in lipid and iron metabolism, and it enhances the biosynthesis of l-carnitine. Due to its antioxidant activity, vitamin C can neutralize the tissue-damaging effects of free radicals. Vitamin C is being related to the prevention of cancer and cardiovascular diseases. This review includes current information on the biosynthesis of ascorbic acid, as new methods are now challenging the traditional Reichstein process for vitamin C’s industrial-scale production. Different strains were analyzed in correlation with their ability to synthesize ascorbic acid, and several separation techniques were investigated for a more effective production of vitamin C.

February 21, 2023 (v1)
Subject: Biosystems
Keywords: DBPs, indicator microorganisms, magnetic field, NaClO, Wastewater
Wastewater disinfection is one of the most critical issues in protecting human health against exposure to waterborne pathogenies. Chlorine is among the most commonly used disinfectants in many wastewaters’ treatment plants. Nevertheless, disquiets regarding chlorine’s disinfection by-products (DBPs) have grown recently. One of the most effective ways to reduce DBPs generation is to reduce chlorine dosage by increasing disinfectant efficiency. Using magnetic field (MF) in wastewater treatment is one of the promising research topics with significant progression. This study aimed to evaluate the efficiency of using a magnetic field and/or sodium hypochlorite (NaClO) disinfection on secondary slaughterhouse wastewater effluent quality and by-products. Three groups of secondary slaughterhouse wastewater effluents were used: G1 was treated with NaClO only at 0, 2, 4, and 6 mg/L; G2 was treated with exposure to MF at 14,500 gausses, and G3 was pretreated with MF, then NaClO at the exact chlor... TGF-β1 and Mechanical-Stretch Induction of Lysyl-Oxidase and Matrix-Metalloproteinase Expression in Synovial Fibroblasts Requires NF-κB Pathways
February 21, 2023 (v1)
Subject: Biosystems
Keywords: LOXs, mechanical stretch, MMPs, NF-κB, TGF-β1
The imbalance in the expression of matrix metalloproteinases (MMPs) and lysyl oxidases (LOXs) in synovial fibroblasts (SFs) caused by mechanical injury and inflammatory response prevents injured anterior cruciate ligaments (ACLs) from self-healing. However, research on the effect of growth factors on SFs on regulating the microenvironment is limited. In this study, mechanical injury and exogenous transform growth factor-β1 (TGF-β1) were employed to mimic a joint-cavity microenvironment with ACL trauma. The function of the NF-κB transcription factor was further studied. The study found that the gene expression of LOXs (except LOXL-1), MMP-1, -2, and -3 in SFs was promoted by the combination of injurious mechanical stretching and TGF-β1 and that the upregulation of MMPs was higher than that of LOXs. In addition, MMP-2 activity induced by the combination of injurious stretch and TGF-β1 was inhibited by NF-κB inhibitors such as Bay11-7082 and Bay11-7085. The findings concluded that the syn... Banana Waste-to-Energy Valorization by Microbial Fuel Cell Coupled with Anaerobic Digestion
February 21, 2023 (v1)
Subject: Biosystems
Keywords: banana waste, biochemical methane potential, bioelectricity, ecofriendly bioenergy, sustainable processes
Banana is the most cultivated fruit plant in the world. It is produced in Latin America, Asia and Africa. India and China are the world’s largest banana producers, with almost 41% of the world’s production. This fruit reaches a total world production of 158.3 million tons per year. However, during their production cycle, the banana agroindustry produces large volumes of solid waste derived from overripe fruit. It contributes between 8−20 percent of the waste (around 100 kg of banana waste for every ton of banana produced). Therefore, the use of overripe banana waste represents a huge opportunity for bioenergy production. This work demonstrates that banana waste can be further used for power generation using a microbial fuel cell (MFC) coupled with anaerobic digestion (AD). First, the maximum methane production (MMP), methane production rate (MPR) and biochemical methane potential (BMP) were measured using an anaerobic batch bioreactor for 64 days of monitoring. Finally, the digestate g... [more]

February 21, 2023 (v1)
Subject: Biosystems
Keywords: electricity generation, high nitrate, microbial fuel cell, nitrate removal, saline denitration solution
Oxidation denitration is one of the most efficient ways to remove NOx from flue gas in a coal-fired power plant. However, this oxidation denitration produces saline solution containing a high concentration of nitrate, which needs to be well treated. In this paper, MFC was firstly used to treat the high nitrate content saline denitration solution from ozone oxidation denitration of a coal-fired power plant. The influences of chemical oxygen demand (COD) and initial nitrate concentration on the nitrate removal and electricity generation of MFC were investigated by sequencing batch mode. The results showed that using MFCs could efficiently remove nitrate from coal-fired power plant saline denitration solution with nitrate nitrogen (NO3−-N) concentration up to 1510 mg/L. The average nitrate nitrogen removal rate was as high as 248.3 mg/(L·h) at initial nitrate nitrogen concentration of 745 mg/L and COD concentration of 6.5 g/L, which was eight times as high as that of the conventional biol... Development of Antibody-like Proteins Targeting the Oncogenic Ser/Thr Protein Phosphatase PPM1D
February 21, 2023 (v1)
Subject: Biosystems
Keywords: adnectin, anti-cancer drug, phage display library, PPM1D, protein phosphatase
PPM1D, a protein Ser/Thr phosphatase, is overexpressed in various cancers and functions as an oncogenic protein by inactivating the p53 pathway. Therefore, molecules that bind PPM1D are expected to be useful anti-cancer agents. In this study, we constructed a phage display library based on the antibody-like small molecule protein adnectin and screened for PPM1D-specific binding molecules. We identified two adnectins, PMDB-1 and PMD-24, that bind PPM1D specific B-loop and PPM1D430 as targets, respectively. Specificity analyses of these recombinant proteins using other Ser/Thr protein phosphatases showed that these molecules bind to only PPM1D. Expression of PMDB-1 in breast cancer-derived MCF-7 cells overexpressing endogenous PPM1D stabilized p53, indicating that PMDB-1 functions as an inhibitor of PPM1D. Furthermore, MTT assay exhibited that MCF-7 cells expressing PMDB-1 showed inhibition of cell proliferation. These data suggest that the adnectin PMDB-1 identified in this study can be... [more]

Subject: Biosystems
Keywords: circular economy, complex, iron, peptides, Saccharomyces cerevisiae, supplementation, yeast
Given the importance of iron in human nutrition and the significance of waste and by-product valorisation in a circular economy environment, we investigated the effects of protein and iron concentration on the production yield of iron−peptide complexes from spent Saccharomyces cerevisiae. For this purpose, different amounts of protein and iron were used in the complexation process. The results have shown that higher concentrations, although permitting a faster and larger scale process, provide a significantly lower complexation yield, which deems the process less feasible. This is corroborated by fluorescence analysis, which shows a lower degree of complexation with higher protein concentration. In addition, varying the concentration of iron does not change the quality of formed complexes, as evidenced by Fourier transform infrared spectroscopy (FT-IR) analysis. The morphology of all samples was also evaluated using scanning electron microscopy (SEM). Therefore, further studies are nee... [more]
